Efficient DNA extraction from young seedlings of cross-pollinating species like buckwheat and lupin enables high-throughput genotyping. To capture genome-wide variability for mapping agronomic traits, at least 100 individuals per cultivar are required. Sowing 150 seeds per variety ensures sufficient seedlings for DNA extraction and phenotyping, streamlining sample collection for GWAFF or DArTseq analysis.
