Sampling Root Exudates and Rhizosphere Soil Using Soil-Hydroponic Hybrid Method

Sampling root exudates and rhizosphere soil using a soil-hydroponic hybrid method in pots, rhizoboxes, or field conditions with barley as an example. The technique involves gentle soil removal from roots, sequential rinsing in fresh solutions to recover rhizosphere material, and controlled exudate collection over 2–3 hours in darkness to minimize disturbance. Roots are rinsed before final immersion in sampling solution to ensure accurate measurement of exudates released due to physiological adjustments. The method preserves root integrity and enables simultaneous collection of rhizosphere soil and exudates.
