In this project, LAMP-based assays were developed and validated for rapid detection of *Pectobacterium carotovorum* subsp. *brasiliense* and *Pectobacterium parmentieri* in potato crops. Primer sets were designed using BLAST analysis to ensure specificity, with performance dependent on DNA concentration. Sample lysis in the lab caused DNA degradation during transport, highlighting the need for on-site lysis to preserve integrity. Positive controls confirmed assay functionality, but variable fluorescence signals indicated challenges with low DNA concentrations or primer sensitivity. FTA paper was tested for sample preservation but yielded inconclusive results. Recommendations include optimizing primer sets and developing reliable on-site lysis protocols to improve field detection accuracy.
